One of the nuns tells Stick (I believe) that Matt has no family except his mom and implies she's a mess of somesort. I don't know if Matt knows his mother is alive or not, though.
Also, doesn't Jack call Matt's mother before the fight with Creel? He calls someone female (they never say who) and leaves a message. He tells her to take care of Matt because he will need her now. And it looks like that doesn't happen, for whatever reason. Let me go and look up the actual dialogue:
"I'm about to go do something. Well, I'm about to go be me. You know better than anybody that doesn't always go so well. I don't know how this is gonna go, but if I were a betting man... Matty's gonna need you. More than ever. Look after him, okay? And I know what I'm asking here. He's a good kid. He sure as hell didn't get that from me, so... It's better this way. Just once, I want Matty to hear people cheer for his old man. Just once..."
In my headcanon, TVshow!Matt doesn't know that his mum is alive. If, like in the comics, she walked out before he could remember her, his dad could have simply kept that fact from him to protect him from the disappointment of having a mother who didn't want him.
SAYRT: You're right, it could be someone other than his mum. I guess my mind just went there straight away, but it's also not unrealistic that it's someone else. Though I have to say from the answering machine text we hear at the beginning, it very much did not sound like it was a nun.
I was about to say that, too, that the way she answered the phone didn't come off like any nun I know. I did think Jack was calling Matt's mom because of the way he tells her, 'He's a good kid. He didn't get that from me' inferring it was from his mother's side of the family. Maybe the MCU is going to give Matt an honest-to-god mom who isn't a nun? That would be weird but so far the MCU hasn't mentioned Karen's father is a villain yet, and over on Agents of SHIELD they did show that Skye's father (Skye who also came from St Agnes orphanage) was the supervillain Mister Hyde (whom DD has fought in the comics). Now I'm going to be wondering if the MCU is going to try to link Matt to an existing superhero who would be where Matt got his 'good' side from.
